Fertilization fo Zona-free Oocyte and Embryo Growth after Microencapsulation

Abstract
The acrosome reaction of sperm in zona pelluicida is found to be not necessary in fertilization process. Many studies about the fertilization of zona-free oocyte revealed considerable fertilization rate in rodent. Recently, after artificial
encapsulation of embryo, the live off-spring could be obtained in rodent model. In this study, we tried fertilization of zona-free mouse oocyte and culture of encasulated embryo to assess the possibility of clinical application of these
procedure.
The fertilization was tried after removal of zona by acidic solution. The zona-free embryo was encapsulated with 1.1% sodium alginate.
The fertilization rate of zona-free oocyte was lower than that of the zona-intact oocyte or zona-intact oocyte with cumulus cell(29.2% vs. 62.3% or 66.7% respectively). The embryo development rates of zona-free embryos with or without sodium
alginate
capsule were significantly lower than zona-intact embryos with or without artificial capsule(22.5% and 44.6% vs. 68.2% and 73.4%).
Form this data we can conclude that the fertilization & embryo growth rate of zona-free oocyte are lower than zona-intact oocyte. And the embryo growth rate after encapsulation with sodium alginate was not significantly affected. But we could not
find
any beneficial effect of encapsulation during the in vitro culture of the embryo.
